Multiphoton fluorescence and second-harmonic-generation microscopy for imaging structural alterations in corneal scar tissue in penetrating full-thickness wound.

چکیده
The transparency of the cornea relies on the special spatial arrangement of stromal collagen. Any pathological abnormality that leads to the wound healing responses and consequential disruption of collagen alignment may hinder corneal transparency. Understanding wound healing may be crucial for the successful applications of clinical procedures such as refractive surgery. Therefore, the development of a noninvasive in vivo imaging technique is valuable for investigating the physiological response associated with corneal wound healing. In this article, we demonstrate multiphoton ex vivo imaging of full-thickness corneal scar tissue 10 years following wounding. We intend to demonstrate the ability of multiphoton imaging to reveal corneal structural alterations from the wound healing process and the potential of this imaging modality in clinical diagnosis and monitoring of corneal pathological abnormalities.

Multiphoton fluorescence and second harmonic generation imaging of the structural alterations in keratoconus ex vivo.
PURPOSE To demonstrate the application of multiphoton fluorescence (MF) and second harmonic generation (SHG) microscopy for ex vivo characterization of the structural alterations of human corneas due to keratoconus. METHODS Buttons of keratoconic corneas derived from penetrating keratoplasty were sent for structural analysis with a custom-built multiphoton microscope. Fluorescence detected wi...
متن کاملSecond harmonic generation imaging of corneal stroma after infection by Pseudomonas aeruginosa
Pseudomonas aeruginosa is a pathogenic gram-negative organism that has the ability to cause blinding corneal infections following trauma and during contact lens wear. In this study, we investigated the directional movement and orientation of an invasive corneal isolate of P. aeruginosa in the corneal stroma during infection of ex vivo and in vivo rabbit corneas using multiphoton fluorescence an...
متن کاملImaging wound healing using optical coherence tomography and multiphoton microscopy in an in vitro skin-equivalent tissue model.
Laser thermal injury and subsequent wound healing in organotypic, skin-equivalent tissue models were monitored using optical coherence tomography (OCT), multiphoton microscopy (MPM), and histopathology. The in vitro skin-equivalent raft tissue model was composed of dermis with type I collagen and fibroblast cells and epidermis of differentiated keratinocytes. Noninvasive optical imaging techniq...
متن کاملImaging coronary artery microstructure using second-harmonic and two-photon fluorescence microscopy.
The microstructural basis for the mechanical properties of blood vessels has not been directly determined because of the lack of a nondestructive method that yields a three-dimensional view of these vascular wall constituents. Here, we demonstrate that multiphoton microscopy can be used to visualize the microstructural basis of blood vessel mechanical properties, by combining mechanical testing...
متن کاملIntegrated structural and functional optical imaging combining spectral-domain optical coherence and multiphoton microscopy
An integrated microscope that combines different optical techniques for simultaneous imaging is demonstrated. The microscope enables spectral-domain optical coherence microscopy based on optical backscatter, and multiphoton microscopy for the detection of two-photon fluorescence and second harmonic generation signals. The unique configuration of this integrated microscope allows for the simulta...
